MOA
1. Reagent methanol
n-Butanol hydrochloric acid solution: Dissolve 5.0 ml of concentrated hydrochloric acid in 95.0 ml of n-butanol, mix well, and store in a cool place.
Ferric ammonium sulfate hydrochloric acid solution: Weigh 2.0 g of NH4Fe(SO4)2.12H2O and dissolve it in 100 ml of 2.0 mol/L hydrochloric acid aqueous solution.

4. Sample solution to be tested: Weigh 25.0 mg of the sample to be tested, dissolve in methanol and dilute to 250 ml.
5. Determination steps:
In a series of 10 ml stoppered borosilicate scale test tubes, add 0.2 ml of ferric ammonium sulfate hydrochloric acid solution, 6.0 ml of n-butanol hydrochloric acid solution, and then add 1 ml of the sample solution to be tested, 1 ml of methanol (blank) , three test tubes were repeated for each treatment. Stoppered and vortexed to mix.
React in a boiling water bath (be careful to ensure boiling water bath) for 40 minutes, take out, quickly cool to room temperature, and record the final volume of the solution in the test tube.
The absorbance relative to the blank was measured with a UV spectrophotometer at 546 nm.
6. Calculation
The percentage of Pure Grape Seed Extract is calculated according to the formula:
Percentage content of proanthocyanidins=A×V/(0.01389×0.366×7.2×100M)×100%
where:
A is the absorbance of the sample to be tested;
M is the weighed amount of the sample to be tested (mg);
The final volume of liquid in the V-tube (ml).
