● Tricarboxylic acid cycle

Alpha-ketoglutaric acid is an important metabolic intermediate in the microbial tricarboxylic acid cycle, serving as a key node connecting the metabolism of carbon and nitrogen in the cell. Its position in the cycle is after isocitrate and before succinyl-coA. At this site, the replenishment of this intermediate metabolite can be achieved by the self-transamination of glutamate to produce α-ketoglutaric acid, as well as by the action of glutamate dehydrogenase on glutamate.

 

● Making amino acids

Glutamate reacts with glutamine synthase, utilizing a molecule of ATP to produce glutamyl phosphate. This intermediate is then attacked by ammonia as a nucleophile to form glutamine and inorganic phosphoric acid.

 

● Dietary supplements

Alpha-ketoglutaric acid powder is sold as a dietary supplement, known as AKG or A-KG, to fitness enthusiasts or bodybuilders who claim it can improve performance during peak training. Sellers claim this is based on research showing that excess ammonia can bind to α-ketoglutaric acid and be carried away to reduce the many problems caused by ammonia poisoning. However, studies demonstrating the effectiveness of alpha-ketoglutarate in reducing ammonia poisoning have only been conducted in patients already undergoing hemodialysis.

Physiological Action

α-ketoglutaric acid is a type of multi-action short-chain carboxylic acid that shows no toxicity and exhibits high stability in aqueous solutions. In vivo, AKG serves not only as an important transitional product in the TCA cycle but also as a precursor of L-glutamate, L-glutamine, L-proline, and L-arginine. Experiments conducted on animals showed that AKG, glutamine, and glutamate ingested by animals were absorbed and metabolized in the upper part of the small intestine, where up to 95% of glutamate and 70% of glutamine were degraded into CO2, while only 40% of AKG was degraded. The remaining AKG can be utilized for metabolic processes in intestinal cells and peripheral tissues.

 

Production Method

● Mix 225 g of triethyl oxalosuccinate with 600 mL of concentrated hydrochloric acid and place the mixture in a container. Distill and concentrate the liquid to 140℃ to obtain 110-112 g of α-ketoylglutaric acid. The yield is 92-93%.

Triethyl Oxalosuccinate: Add 360 mL of anhydrous ethanol to a reaction vessel equipped with an agitator and reflux condenser. Gradually add 23 g of clean sodium metal in batches. After addition, heat the mixture to reflux to complete the reaction with the sodium metal. Transfer to a distillation apparatus and distill off the ethanol. Simultaneously, continue adding toluene until the internal temperature reaches 105℃. Cool the mixture to room temperature, then add 650 mL of anhydrous ether. Stir in 146 g of diethyl oxalate and then add 174 g of diethyl succinate (1.0 mol). Let the mixture stand at room temperature for 12 hours. After stirring, add 500 mL of water to separate the organic layer, which is then extracted with 150 mL of water. Combine the water layers, acidify with concentrated hydrochloric acid, and precipitate the oily substance. Separate the oil layer and extract the water layer with ether three times. Combine the organic layers and dry with anhydrous magnesium sulfate.

 

Recover the ether to obtain 235 to 250 g of yellow oil compound with a yield of 86% to 91%. α-Keto-glutaric Acid: In a reaction vessel equipped with a blender and reflux condenser, add 225 g of the above compound, 330 mL of concentrated hydrochloric acid, and 660 mL of water. Stir the mixture and reflux for 4 hours. Concentrate under reduced pressure to dryness (60-70 ℃). Add 200 mL of nitroethane to the residue, heat to dissolve, and filter while hot. Combine the nitroethane solution, then stir the mixture at 0-10 ℃ for 5 hours. After filtration and drying at 90℃ for 4 hours, obtain 88-99 g of compound with a yield of 73%-83%.
