I3C has been used in the treatment of patients with recurrent respiratory multivessel papilloma. In a prospective study, 24 adult patients (200 mg) and 9 pediatric patients (3 mg/kg) were given I3C at dose dependent doses, all twice daily. The results showed that 10 adult patients and 1 pediatric patient did not require further surgical treatment for 31 to 75 months (mean 56.7 months) after taking I3C. 7 adult patients and 3 pediatric patients required less frequent surgical treatment for 41 to 76 months (mean 56.2 months) thereafter; another 7 adult patients and 5 pediatric patients did not respond significantly to I3C treatment.
No immediate or long-term side effects were observed throughout. And the mechanism of the difference in the efficacy of I3C in patients with recurrent respiratory multivessel papilloma may be related to differences in estrogen metabolism.
Moreover, the results of this study suggest that pure Indole-3-methanol powder is more effective in adult patients and is not guaranteed to produce definitive efficacy in pediatric patients. Similar to this study, an 8-year-old female patient with recurrent respiratory multiforme papilloma treated with an anti-human papillomavirus drug of established efficacy along with adjuvant therapy with I30 resulted in complete resolution of the pulmonary infection by a combination of internal and intravenous injections into the lung infection site, and the effect lasted for at least 24 months.
Phase I clinical trials3 of the chemopreventive effect of I3C on breast cancer noted that 3C could significantly increase 2-0HE levels and decrease 16a-0H E levels, thus exerting an anti-breast cancer effect. Women with systemic lupus erythematosus (SLE) have an estrogen metabolism that tends to produce 16a-OH estrone, which may promote disease development. Since I3C can alter the direction of estrogen metabolism, it can be metabolized to produce substances with weaker estrogenic activity.
Therefore, McAlindon et al26 studied the effect of 375 mg of I3C daily in women with SLE and showed an increase in the urinary 2-0H estrone/16a-OH estrone ratio and a decrease in the SLE activity index, suggesting that this effect may be due to the anti-estrogenic effect of I3C.
