Standard test procedure

● Description:

Light yellowish to brown powder

 

● Identification :

The retention time of the Andrographolide peak in the chromatogram of the assay preparation corresponds to that of theAndrographolide peak in the chromatogram of the Standard preparation as obtained in assay.

ASSAY (Andrographolide CONTENT):

By High Performance Liquid Chromatography.

● Standard preparation:  

Take an appropriate amount of andrographolide powder reference substance, accurately weigh it, add methanol to make a solution containing 0.1 mg per 1 ml, and get it.

 

● Sample preparation:

Take about 0.05 g of the test sample, accurately weigh 500ml volumetric bottle, add 95% ethanol 300 ml, ultrasonic for 10 minutes to dissolve, add water to scale, then ultrasonic for 30 minutes, add water to scale after room temperature, shake well, then get.

 

● Procedure:

Separately inject equal volume (20µl) of the standard preparation and the test preparation into chromatogram in the triplicate and note down the area counts of Andrographolide peak calculate the percentage of Andrographolide.

 

Calculation:

           

 At and As are the counts of Andrographolide peak in the chromatograms of the test preparation and standard preparation respectively. Ps is the contents of Andrographolide in standard. 

 

Properties:

● Appearance:

white square prismatic or flaky crystals (ethanol or methanol), odourless, bitter taste. Powdered white powder, 80 mesh.
●Solubility:
Andrographolide powder can soluble in boiling ethanol, slightly soluble in methanol or ethanol, very slightly soluble in chloroform, almost insoluble in water or ether.
●Melting point:
230～231°C.
●Spinning luminosity:
(α)17D -126.6±2° (glacial acetic acid), [α]D -112.7 (c,0.53, MeOH).

●Chromogenic reaction:
With basic 2% 3,5-dinitrobenzoic acid reagent, shows purple-red colour; reacts positively with iron isohydroxamic acid.
●Density:
(21°C) 1.2317g/cm3.

●IR:

Liquid phase detection methods

Octadecylsilane-bonded silica gel was used as filler. Methanol-water (60:40) was used as the mobile phase. The detection wavelength was 225nm.

 

●Preparation of control solution
Appropriate amount of control was taken and weighed precisely. Then add methanol to make a solution containing 0.1 mg per 1 ml.

●Preparation of test solution
Take about 25mg andrographolide powder and weigh it precisely. Then put it in a 50ml measuring flask, add methanol to dissolve and dilute it to the scale, shake well. Measure 5ml, put in 25ml measuring flask, dilute with methanol to scale, shake well, that is to obtain.

●Determination method:
Take up the control solution and test solution 10μl respectively. Then inject into the liquid chromatograph and determination.
