Brief Introduction
Curcumin extract powder is a diketone compound extracted from the rhizome of some plants in the ginger family and Tennantaceae. Turmeric contains 0.3-5.4% curcumin, an orange yellow volatile oil composed mainly of turmerone,atlantone and zingiberone. Curcumin provides 95% Curcuminoids. Curcumin is orange-yellow crystalline powder, slightly bitter, insoluble in water and ether, soluble in ethanol, propylene glycol, easily soluble in glacial acetic acid and alkaline solution, reddish-brown in alkaline, yellow in neutral and acidic. Curcumin content(HPLC) determination method
Summary of methodology
After the sample was dissolved by ultrasound, it was determined by HPLC and quantified by external standard method.
II. Instruments and tools

High performance liquid chromatography (with ultraviolet detector)
III. Reagents and solutions
① Acetonitrile. Chromatographic purity;
②Methanol; chromatographic purity;
③ glacial acetic acid; chromatographic purity;
④ HPLC grade water or MILIQ water;
4. Reference substance of curcumin: purity of HPLC ± 98%.
Five Chromatographic Conditions and System Applicability
5.1 CHROMATOGRAPHIC CONDITIONS
A) Chromatographic column: octadecylsilane bonded silica gel column, 4.6 mm X 250 mm.
B) mobile phase: acetonitrile-4% glacial acetic acid solution (48:52), filtered by an organic system 0.45 um filter membrane, and prepared by ultrasonic.
C) Flow rate: 1.00 ml/min.
D) detection wavelength: 430 nm.
E) Column temperature: 30 ℃
5.2 System serviceability
The theoretical plate number should not be less than 4000 according to the calculation of curcumin peak.
VI. Methods of operation
6.1 Sampling
The samples were sampled according to the method in Appendix II A of the 2010 edition of the Pharmacopoeia of the People's Republic of China.
6.2 Preparation of Control Solution
The reference substance of curcumin (about 10 mg) was weighed accurately, placed in a 100 mL brown bottle, ultrasonic dissolved with methanol and diluted to a scale to obtain a reference substance solution of 0.10 mg/ml.
6.3 Preparation of solutions for test products
Take an appropriate amount of turmeric extract powder (according to the content), accurately weigh, put into a 100mL brown bottle, add about 80mL methanol, extract for 15 minutes by ultrasonic, take out, put into room temperature, dilute with methanol to the scale, shake well.The test solution was obtained by filtering the filtrate with organic 0.45 um microporous filter membrane and placing the filtrate in a brown sample bottle.
6.4 Methods of measurement
The reference solution and the sample solution were precisely absorbed and injected into the liquid chromatograph respectively, and the content was determined by external standard method.
Note: The chemical property of curcumin is not stable. The control and the sample solution should be detected within 2 hours after preparation.
7 Result Calculation
The content is calculated according to formula (C1):

Applications
● Food additives
Turmeric extract has long been widely used in the food industry as a commonly used natural pigment, mainly in the coloring of canned food, intestinal products and sauce and marinade products, and the amount used depends on the normal production needs. The product form of functional food with curcumin as the main ingredient can be general food or some non-food forms, such as capsules, pills or tablets. For the general food form, some yellow vegetarian food products can be considered, such as pastries, sweets, beverages, etc.
Curcumin is a food additive approved by the Codex Alimentarius Commission of the Food and Agriculture Organization of the United Nations (FAO/WHO-1995), and is one of the first nine natural pigments allowed to be used in food as promulgated in China's Hygienic Standard for the Use of Food Additives.

Used as an acid-base indicator, pH 7.8 (yellow)-9.2 (red-brown).
● Medicine
Turmeric is commonly used as medicine, and its main bioactive components are curcumin-like and volatile oil. The former has hypolipidemic, anticoagulant, antioxidant and choleretic effects, while the latter mainly acts as an anti-inflammatory, antibacterial and cough suppressant.
