Extraction methods:

Our products use Grain alcohol / water and methods. Let me introduce these two methods.

1. Water Process 

● Rice bran is cleaned and then soaked in water to soften the bran.

● The softened rice bran is then cooked in boiling water for a set amount of time to release the beneficial compounds.

● The mixture of rice bran and water is then separated using a filter or other separation method.

● The extracted liquid is then further processed and dried to obtain the desired rice bran extract powder.

The water process is considered a natural and environmentally friendly method for extracting the valuable nutrients from rice bran, with minimal chemical or solvent use.

2. Grain alcohol

● Extraction: 

Rice bran is cleaned and extracted with high proof grain alcohol, usually ethanol or methanol, to extract the bioactive compounds.

● Filtration: 

The extract is then filtered to remove any impurities or solid particles.

● Concentration: 

The extract is concentrated by evaporating the solvent under vacuum to obtain a concentrated solution.

● Drying: 

The concentrated solution is then dried using spray drying or freeze drying to obtain a dry powder.

● Packaging: 

The dried powder is packed in airtight containers and stored in a cool, dry place. 

HPLC Process:

● Sample Preparation: 

a. Weigh accurately 1gm of rice bran extract powder and dissolve it in 5ml of methanol.

b. Filter the solution through a 0.45-micron membrane filter.

c. Collect the filtrate into a vial to be used for HPLC analysis.

● Standard Preparation:

a. Prepare stock solutions of reference standards (e.g., ferulic acid, gamma-oryzanol, etc.) of known concentrations in methanol.

b. Dilute the stock solutions to obtain a working solution of a known concentration of each standard.

● HPLC Conditions:

a. Use an HPLC instrument equipped with a C18 reverse-phase column and a photodiode array detector.

b. Set the wavelength of the detector at 320 nm.

c. The mobile phase should consist of a mixture of (A) water containing 0.1% formic acid and (B) acetonitrile with 0.1% formic acid.

d. The flow rate of the mobile phase should be maintained at 0.8 ml/min.

e. The injection volume of the sample should be 10 µL.

● Calibration Curve: 

a. Inject the working standard solutions of ferulic acid, gamma-oryzanol, etc. in triplicate into the HPLC instrument under the described chromatographic conditions.

b. Record the peak areas for each standard and plot a calibration curve of peak area versus concentration.

● Sample Analysis:

a. Inject the prepared sample solution into the HPLC instrument under the described chromatographic conditions.

b. Record the peak areas for each compound and calculate the concentration of each compound using the calibration curve.

● Report:

a. Report the concentrations of each compound present in the analyzed sample.

b. Present the results as mean ± SD of the triplicate analysis.
