Production Method For Hesperidin and Hesperetin from Orange Peel

This document outlines two methods for extracting bioactive compounds from dried orange peels. The primary compounds are Citrus Aurantium Extract Powder hesperidin, a glycoside, and hesperetin, its aglycone counterpart.

Method 1: Production of Hesperidin Powder

Citrus Aurantium Extract Powder is naturally found in the peels of citrus fruits like oranges and lemons. This method describes its extraction from dried, ripe orange peels.

●Preparation:

First, the dried orange peel is coarsely chopped. It is then soaked in 3 to 6 times its weight in water for 30 minutes to soften.

●Alkaline Extraction:

To the softened peel, 4-10% lime (calcium hydroxide) and 7-12 times the amount of water are added. The mixture is stirred, and the pH is checked and adjusted to a highly alkaline range of 11.5 to 12.0 using lime or sodium hydroxide. It soaks for 1.5 to 2 hours.

●Filtration and Washing:

The mixture is centrifugally filtered. The solid filter residue is washed again with 5-7 times its weight in water, and the pH is readjusted to 11.5-12 for a second soaking and filtration. This repeats until the filtrate is clear.

●Precipitation: The combined clear filtrate is acidified to pH 5 using dilute hydrochloric acid. After standing for two days, a precipitate forms.

●Purification and Drying:

The precipitate is collected and washed with water until the solution is nearly neutral. This is the crude hesperidin. To purify it, the crude product is dissolved in a solution of 1% sodium hydroxide in 50% ethanol and filtered. The filtrate is re-acidified to pH 5 with hydrochloric acid to precipitate the pure compound. The final precipitate is washed, dried at 70°C, crushed, and sieved into a powder. The total yield is 0.6% to 1.8% of the original orange peel powder weight.

 

Method 2: Production of Hesperetin

Hesperetin is derived from Citrus Aurantium Extract Powder hesperidin through a process that breaks down its structure.

 

●Alkaline Hydrolysis:

Coarse orange peel powder is mixed with water and lime. The pH is adjusted to a very high range of 12 to 13 using a 25% sodium hydroxide solution. This mixture soaks for 10 to 12 hours.

Filtration: The liquid is pressed out and collected, while the solid residue is discarded.

●Acidification and Heating:

The pH of the liquid filtrate is lowered to 3-4 using concentrated hydrochloric acid. This acidic solution is then heated and maintained at 70°C for 30 minutes.

●Precipitation and Drying:

The solution is cooled and allowed to stand until precipitation is complete. The solid precipitate is collected via suction filtration, washed with water until neutral, and dried at 80°C. This results in crude hesperetin.

●Purification:

Citrus Aurantium Extract Powder Pure hesperetin is obtained by recrystallizing the crude product using methanol as a solvent.
