Effects of Epimedium Extract Powder on Immunity and Its Potential Applications

● The Role and Mechanism of Immune Regulation - Lymphocytes

Th1/Th17 cells: Th17 is closely related to Th1 during development, and overexpression of these cells is associated with many inflammatory diseases. ICA regulates lymphocyte function by influencing the Th1/Th17 and Th2 balance. In a collagen II-induced arthritis (CIA) model, ICA treatment reduced the proportion of CD4+IL-17+ cells and the number of Th17 cells. Serum levels of IgG2a also decreased, along with reduced arthritis scores. However, additional IL-17 administration negated these effects [16], suggesting ICA primarily controls CIA by mediating Th17 cells.

In an experimental autoimmune encephalomyelitis (EAE) model, ICA-treated mice showed a reduced frequency of Th1 and Th17 cells in the spleen and lymph nodes. Fewer Th17 cells were also observed in CNS monocytes of ICA-treated mice. Interestingly, some studies suggest Epimedium extract powder may enhance immune responses, as it stimulates Th1 lineage development in mice and promotes IgG production.

● Inflammatory Cytokines

ICA exerts multiple effects on inflammatory cytokines in different diseases. In bronchoalveolar lavage fluid (BALF) cell cultures, ICA significantly inhibited interleukin-6 (IL-6) and IL-17 but did not affect IL-10 levels.

In rats with LPS-induced brain dysfunction, ICA significantly improved spatial learning and memory while reducing TNF-α, IL-1β, and COX-2 expression in the hippocampus. In titanium-stimulated mice, ICA reduced TNF-α, IL-6, and IL-1β secretion in the calvaria. ICA also inhibited the gene expression of IL-1β, IL-8, TACR1, and ICAM-1 in HaCaT cells in a dose- and time-dependent manner.

However, in another in vitro pharmacology study on concanavalin A (ConA)-stimulated T lymphocytes, ICA decreased IL-2, IL-4, and IL-10 production while upregulating TNF-α and IFN-γ. These findings suggest ICA's immune-modulating effects may vary depending on the microenvironment.

RORγ: RORγ is a key transcription factor controlling the function and development of CD4+ Th17 and CD8+ Tc17 cells. RORγ enhances Th17 function by increasing cytokine and chemokine secretion, such as GM-CSF and IL-17. In an ovalbumin (OVA)-induced asthma model, ICA reduced the number of CD4+ RORγ+ T cells, significantly lowering RORγ levels.

T-bet: The T-box factor (T-bet) is a "master regulator" of Th1 cells and plays a crucial role in immune responses. In ICA-administered asthmatic rats, T-bet and GATA-3 mRNA expression were reduced, helping to restore Th1/Th2 balance.

In a rat model of unpredictable chronic mild stress (CMS)-induced depression, ICA administration for 35 days alleviated depressive behaviors and increased GR mRNA expression. In EAE mice, ICA exhibited estrogen-like activity, modulating HPA function and increasing white matter GR expression.
