Jerusalem Artichoke Inulin Powder is a white powder. It is an indigestible plant fructan found in members of the Compositae family. Commercial production is by extraction from the roots of Cichorium intybus Lin. (chicory). It is a mixture composed of polymers of β-2,1-linked fructose residues mostly linked to a terminal glucose residue. The degree of polymerization in the mixture varies between 3 and 60, with the longer-chain polymers being predominant. It is very soluble in hot water, slightly soluble in cold water, and almost insoluble in most organic solvents.

Assay- Procedure

● Mobile phase: Water

● Standard solution: 5 mg/mL fructose, 1 mg/mL glucose, and 1 mg/mL sucrose

● Sample solution: Transfer 1 to 1.5 g of Jerusalem Artichoke Inulin Powder sample into a 100-mL volumetric flask. Dissolve the Jerusalem Artichoke Inulin Powder sample in hot (> 80°) water and allow the solution to cool before diluting to volume.

● Mode: High-performance liquid chromatography

● Detector: Refractive index

● Column: Gel filtration column for molecular weight range up to 5000 Da (Shodex KS-802, Showa Denko, K.K., Tokyo, Japan, or equivalent)

● Temperature Column: 50°

● Detector: 35°–40°

● Flow rate: 1 mL/min

● Injection volume: 20 μL Sample loop: 20 μL

● Analysis: Separately inject the Standard solution and the Jerusalem Artichoke Inulin Powder Sample solution into the chromatograph and record the chromatograms. Determine the percentage of each sugar and of Jerusalem Artichoke Inulin Powder in the Sample solution using the following formula:

 

Result = 100(CST × ASA)/(AST × W)

CST  = concentration of each sugar in the Standard solution (mg/100 mL) 

ASA = area of the corresponding sugar peak in the chromatogram of the Sample solution

AST = area of the corresponding sugar peak in the chromatogram of the Standard solution

W = weight of the sample taken (g/100 mL)
