Analysis Method - Content of Matrine

● Solvent:

A mixture of alcohol and acetonitrile (80:20)

● Solution A

Use a filtered and deaerated solution of 3ml of phosphoric acid in 100ml water

●Mobile phase

Use a filtered and degassed mixture of acetonitrile, alcogol and solution A (80:10:10)for the Chromatographic system.

Using a UV absorption at 220 nm.

 

● Standard solution

Transfer an accurately weighed quantity of standard Sophora Root Extract Matrine, equivalent to about 0.1mg of the labeled content of matrine to a100mlvolumetric flask.Add solvent into the flask.dissolved and mix it.

● Test solution

Accurately weigh approximately 25mg of Liquid Matrine to a 100ml volumetric flask. Add So vent into the flask, dissolve it and then dilute with Solvent to volume ,mix it.

● Procedure

Separately inject equal volumes(20ul)of Standard Solution and Test Solution into the Chromatograph, record the chromatograms, and measure the areas of the analyse peaks. Identify the retention of the peaks corresponding to matrine by comparison of the chromatogram of Standard Solution with that obtained from the Reference Chromatogram. Separately calculate the percentages of Matrine by the formula:

Weigh the appropriate amount of bitter ginseng base control into a 25ml volumetric flask, dissolve it with methanol by ultrasonic and dilute it to the scale. Filter with 0.45μm filter membrane and prepare for use.
