Introduction:

Dextran powder is also known as glucose polymer powder. It is a homotypic extracellular polysaccharide composed entirely of α-D-glucopyranose monomers. It mainly consists of α (1,6) glycosidic bonds connected by first and last dehydration condensation, and also contains different ratios of α (1,2), α (1,3), α (1,4) glycosidic bonds connected by branches. It is not only the world's first industrialized microbial polysaccharide, but also a recognized plasma substitute at home and abroad.
Dextran, which is generally used in clinical practice, contains about 95% of alpha (1,6) chain. With high α (1,6) chain, it degrades slowly in vivo and expands blood volume persistently. However, when the alpha (1,6) chain is nearly 100%, its solubility is poor and it is not suitable for formulation. Immunological studies have shown that clinical side effects are lower when the α (1,6) chain is about 95%.

Properties  
● White amorphous powdery solid. Glucose polymer powder is odorless, tasteless, soluble in water (the smaller the molecular weight, the more soluble in water), insoluble in methanol, ethanol, ether, acetone. Specific spin [α]D = +1900 ～ +2010 (at 25 ℃).
●Dextran is electrically neutral. So there is no isoelectric point or electropherogram. It can be stable at room temperature or in neutral solution, hydrolyzed by heating together with strong acid. And its end group is easily oxidized in alkaline solution. Dextran anhydride solution reacts with sodium hydroxide and copper sulphate solution to produce a light blue precipitate, which turns into brown lumps when heated. It changes color or decomposes when subjected to high temperature, and slight depolymerization can occur when heated in vacuum at 100℃. When heated at 150℃ it will dehydrate and change color, when heated to 210℃, it will decompose completely in 3~4 hours.
● Dextran raw material molecular weight is large and uneven molecular weight distribution will make the preparation dissolution produce more obvious emulsion light, the proportion of large molecules is too large will lead to the difficulty of filtration of drug solution.

● The chemical structure of glucose polymer powder is related to the temperature and concentration of fermentation, when the temperature of the fermentation solution is 18, 23, 30 ℃, the percentage of flaky precipitate in its injection solution is 35%, 14%, 3% respectively. When the temperature of the fermentation broth was 37°C, no flaky precipitate appeared in its injection.
● When the amount of calcium and magnesium ions in the culture medium is increased. It decreases the content of dextran containing a(1,6) chain from 95% to 82%.
● When the composition and proportion of nitrogen source in the medium changes, it also affects the proportion of main chain and secondary chain in the structure of dextran anhydride, resulting in difficulty in filtration of injections, emergence of milky light and poor clarity.
● Traditional process with ethanol division of molecular weight, it is difficult to strictly control the molecular weight distribution range of dextran anhydride. Ethanol concentration is high. Dextran colloidal precipitation is too fast. Impurities are wrapped in the precipitated particles. In addition to denaturation of proteins, attached to the surface of precipitated particles, preventing the elution of other impurities, and ultimately the formation of larger particles or even lumps of sticking to the cans. The raw material particle size is large, poor solubility, yellowish color. If the ethanol concentration is low, the colloid settling speed is slow, water-soluble impurities are not easy to remove, the raw material particle size is small.
