Identification Report

1. Identification Purpose

To confirm that the testing andrographis extract powder is prepared from the raw materila of Andrographis Paniculata., by means of extraction, separation and purification.

 

2. Identification Method

2.1 By TLC

Take 0.05 g Andrographis paniculata extract, add 25 ml of ethanol, sonicate for 30 minutes, filter, and take the supernatant as the test solution. Another 0.5g of Andrographis paniculata raw herb powder was used as a control medicinal solution. Then take andrographolide reference substance, add absolute ethanol to make a solution containing 1mg per 1ml, as a reference solution. According to the thin layer chromatography (general rule 0502) test, draw 1 ul of each of the above three solutions, respectively point on the same silica gel GF254 thin layer plate, using chloroform: ethyl acetate: methanol (4: 3: 0.4) as the developing agent, expand , Take it out, let it dry, and inspect it under an ultraviolet light (254nm). In the chromatogram of the test andrographis extract powder, the fluorescent spots of the same color are displayed at the positions corresponding to the chromatogram of the reference medicinal material and the chromatogram of the reference substance.

● Standard preparation:

Take an appropriate amount of andrographolide reference substance, weigh accurately, add absolute ethanol to make a solution containing 0.1mg per lml.

 

● Test solution preparation:

Take approximately 0.01g of Andrographis Extract Powder, weigh accurately, and place it into a 25ml volumetric flask. Add an appropriate amount of ethanol solution, sonicate for 10 minutes, and then allow the mixture to reach room temperature. Dilute the solution with ethanol, shake, filter, and collect the filtrate. This will yield the extract solution.

Take about 0.5g Andrographis paniculata powder, weigh it accurately, put it in a 50ml volumetric flask, add an appropriate amount of ethanol solution, sonicate for 30 minutes, put it to room temperature, dilute the volume with ethanol solution, shake well, filter, take the filtrate, then get the raw material solution.

Accurately draw 20 μL of the reference solution, the extract solution, and the raw material solution into the HPLC detector, then test.
