Nicotinamide adenine dinucleotide (NAD) is a key coenzyme found in all living cells. It is a dinucleotide consisting of two nucleotides linked by a phosphate group. In one experiment, NMNH (100μM) treatment increased cellular NAD+ levels by 5-fold in HepG2 cells, whereas NMN (100μM) only slightly increased NAD+ levels as determined by mass spectrometry. The results show that NMNH is a more potent NAD+ than NMN. In addition to HepG2 cells, we also confirmed that NMNH can also enhance NAD+ in ES-2 (ovary derived cells) cells. We also found that NMNH increased cellular NAM concentration. These results indicate that NMNH is a more potent NAD+ enhancer than NMN in vivo. NMNH powder also significantly increased NAM levels in the liver, and it was confirmed that high doses of NMNH were not hepatotoxic.
● NMNH inhibited Glycolysis and TCA Cycle
NMNH can reduce the levels of glycolytic intermediates and the levels of TCA cycle intermediates, including intermediates in the glycolytic pathway. These results indicate that accumulation of NADH produced by NMNH inhibits cellular glycolysis and TCA cycle. In contrast, NMN treatment of glycolysis and TCA cycle only induced a slight decrease in products in glycolysis, and almost no reduction in TCA cycle cells was observed.
