● Ultrasound-assisted method
Ultrasound-assisted method is the use of sound waves generated by high-speed, strong cavitation effect and stirring effect, so as to destroy the original material cell structure, accelerate the solvent penetration, shorten the extraction time and improve the extraction rate.
The advantages of this method are: (1) high extraction efficiency; (2) short extraction time; (3) low extraction temperature; (4) wide application; (5) less impurities in the extraction solution, which facilitates separation and purification; (6) lower operating cost of the extraction process; (7) easy operation and convenient equipment maintenance and servicing.
Giacometti et al. used response surface analysis to optimize the ultrasonic-assisted extraction process of lignan-4′-O-glucoside and several other components in olive leaves, and investigated the effects of different ultrasonic operation methods (pulsed and continuous), liquid-solid ratio and ultrasonic time on the yield of each component. Zhong Fangli et al. obtained 2.175 mg/g of lignoceran from the persistent calyx of Gymnema sylvestre using ultrasonication, and Zhang Hongmei selected Gynostemma sylvestre for a similar study and obtained 2.437 mg/g. Cui Jianqiang et al. optimized the extraction of lignoceran from peanut shells by ultrasonication using response surface methodology, and the extraction rate was 0.43% after optimization. Compared with the traditional process, ultrasonic extraction is highly automated and the yield can be increased by 3-4 times, and the domestic ultrasonic extraction equipment with industrial application is now available for industrial production.


●Microwave-assisted method
Microwave-assisted luteolin powder extraction method is a new technology using microwave energy for material extraction. The method is through the appropriate amount of suitable solvent in the microwave reactor from natural plants, minerals or animal tissues to extract a variety of chemical components, with high efficiency, rapid, environmental protection, wide application and other characteristics. The main influencing factors of the process are the choice of solvent, liquid-solid ratio, microwave radiation time, microwave radiation power, the degree of fragmentation, the effect of moisture or humidity in the material, and other factors affecting microwave extraction (intensification).
Wang et al. studied the extraction of lignocaine and apigenin from peony pods by microwave-assisted method, and 151 μg/g lignocaine could be extracted under the optimal process conditions. This method allows the solvent to be heated rapidly during the extraction process, and the cell walls of the plant material are rapidly destroyed to accelerate the dissolution and extraction of the components. Cui Jianqiang et al. used single-factor experiments and response surface method to study the process of using microwave-assisted method to obtain lignocaine from peanut shells, the optimal scheme of microwave time of 70 s, microwave power of 1000 W, lignocaine yield of 0.713%. The same study was done by Jin Shi et al. using lignum vitae leaves. Microwave extraction has a large increase in extraction rate, energy-saving effect, but the microwave extraction needs to be carried out before the herb soaking treatment, the comprehensive extraction time is long, and large microwave extraction equipment still has many shortcomings, the extraction line is difficult to continuous, easy to achieve in experimental studies, industry still needs further research.


● Supercritical fluid extraction method
Supercritical fluid extraction refers to the separation operation of extracting soluble components from solids or liquids with supercritical fluids as solvents. Supercritical fluid has similar diffusion coefficient of gas, solubility of liquid, and surface tension of 0. It can quickly penetrate into the solid material and extract the essence of its components, which has the characteristics of high efficiency, not easy to oxidize, pure natural, and no chemical pollution.
Wang Shengli et al. explored the process conditions of supercritical CO2 extraction of lignocaine from peanut shells by response surface method optimization, and the obtained lignocaine extraction rate increased from 51.2% by ethanol reflux method to 87.8% by supercritical CO2 extraction process.Devequi-Nunes et al. chemically characterized six different propolis extracts by conventional method and supercritical extraction to study their biological activities. The results showed that for obtaining antioxidant compounds with the highest content (containing lignans), supercritical fluid extracts were lower than ethanolic extracts. Although the supercritical method has a faster extraction rate, no solvent residue, and prevents the presence of harmful substances and environmental pollution, the extraction equipment is more expensive and the extraction rate is not significantly improved.
