The HPLC method was used to determine the contents of magnolol and honokiol in magnolia bark supercritical CO2 extract. The extraction process of magnolia bark supercritical CO2 was optimized through an orthogonal test, and the antioxidant activity of magnolia bark extract powder was detected using the MTT method. Under the optimal extraction process conditions, the extraction efficiency of magnolol and honokiol was high, and the process was stable and feasible with good repeatability. The extract showed good antioxidant activity.

Preparation of Reference Substance and Test Solution:

● Preparation of Reference Solution:

Precisely weigh 2.01 mg of magnolol reference substance and 2.32 mg of honokiol reference substance, place them in 25 mL measuring bottles respectively, add methanol to the mark, shake well, and prepare a mixed reference substance solution containing 80.40 μg/mL of magnolol and 92.80 μg/mL of honokiol.

● Preparation of the Test Solution:

Magnolia bark extract were passed through a 40-mesh sieve and dried in an oven at 60°C. The pieces were then put into a barrel and placed in a supercritical CO2 extraction device for extraction. The extracts from separation kettle 1 and 2 were collected and mixed evenly. An appropriate amount of the extract was accurately weighed and placed in a 25 mL measuring bottle. Methanol was added to dissolve it and dilute it to the mark. The solution was sonicated for 5 minutes, allowed to stand, and passed through a 0.45 μm microporous membrane to  obtain the test solution.

Some researchers have discovered that magnolol, the active ingredient in magnolia bark, has a potent inhibitory effect on Streptococcus mutans, a bacterium that causes dental cavities. Additionally, it can effectively eliminate Staphylococcus aureus, with a minimum inhibitory mass concentration of 6.25 μg/mL. Thus, Magnolia officinalis bark extract can be utilized in cosmetics to alleviate skin acne while also serving as a natural preservative.

● Anti-acne and anti-inflammatory

Honokiol, on the other hand, has a potent inhibitory effect on two acne-causing bacteria, P. acnes (Propionibacterium acnes) and P. granulosa (Propionibacterium granulosum), with a minimum inhibitory concentration (MIC) of 9mg/L. Furthermore, it can effectively reduce the rise of IL-8 and TNF-α in THP-1 cells caused by P. acnes, indicating that magnolol can aid in treating local skin inflammation.

● Anti-aging and whitening

Research has demonstrated that magnolia bark extract powder can increase antioxidant enzyme activities like SOD, CAT, and GSH2Px in mouse serum and tissues, reducing the content of lipid peroxidation production (MDA) in the body. The antioxidant capacity of the body is improved under magnolol's influence, and excessive free radicals generated by the aging body can be eliminated quickly, hindering the further chain reaction of oxygen-free radicals. This process inhibits the peroxidation of tissue cells in the body, achieving the goal of anti-aging. Furthermore, the total magnolol's scavenging ability is stronger than that of the classic antioxidant ascorbic acid. Total magnolol can also inhibit tyrosinase activity, providing a certain whitening effect, and may be used as a skin whitening component in cosmetics.
