Introduction:

Diindolylmethane DIM Powder is a molecule named after its structure, where two indole groups are linked to a methane group. Diindolylmethane bulk powder (3,3'-Diindolylmethane) is a white to off-white crystalline powder, soluble in methanol, ethyl acetate, dichloromethane and other organic solvents. It is soluble in alcohol, sealed at low temperature and protected from light. It is widely found in natural plants of the cruciferous family, and has important biological activities. At present, it has been used in the field of pharmaceutical-grade healthcare products.

3.Measuring steps：

Precision weighing 5mg Diindolylmethane standard,dissolve into a 25ml volumetric flask

with acetonitrile.

Precision weighing two samples to be tested 5mg, respectively in a 25ml volumetric

flask,dissolve with acetonitrile;

Ultrasonic respectively for 5min,put at room temperature,sample injection.

Preparation

DIM Powder (1.17g, 10mmol), CTAB (50%mol) and oxalic acid (50%mol) were placed in a 25ml single-neck round bottom flask, 5 ml of deionized water was added, and after fully stirring for 5 minutes, aqueous formaldehyde ( 0.38 g solution, 5 mmol formaldehyde).

 

The reaction was carried out at room temperature for 3 hours, and the reaction was stopped. The reaction solution was extracted three times with 15 ml of ethyl acetate, and the organic phase was collected and dried over anhydrous Na2SO4 for 5 hours. 

 

The solvent was removed under reduced pressure and recrystallized from a mixed solution of methanol and water (methanol/H2O=10/1) to obtain 3,3'-diindolylmethane as a white solid with a yield of 85%.

 

Biological Activity of 3,3′-Diindolylmethane (DIM)

3,3′-Diindolylmethane DIM Powder is a biologically active compound formed during the digestion of indole-3-carbinol, a natural substance found in cruciferous vegetables such as broccoli, cabbage, and Brussels sprouts. Recognized for its promising anticancer and hormone-modulating properties, DIM plays a significant role in regulating cellular processes both in vitro and in vivo.

 

In Vitro Studies

DIM Powder exhibits potent radioprotective and radiomitigation effects in cell-based studies. It activates DNA damage response (DDR)-like mechanisms, primarily driven by ATM (ataxia-telangiectasia mutated) kinase, which plays a central role in maintaining genomic stability. Additionally, DIM stimulates NF-κB survival signaling, enhancing cellular resilience to radiation-induced stress.

As an anticancer agent, DIM has shown the ability to suppress tumor cell proliferation, inhibit invasion and angiogenesis, and induce apoptosis across a variety of cancer cell lines. These effects are mediated through its influence on several key signaling pathways, including AKT, NF-κB, and FOXO3, which are closely associated with cell growth, survival, and programmed cell death.

Moreover, DIM interferes with hormonal signaling, particularly by inhibiting estrogen-induced gene expression. It induces endoplasmic reticulum (ER) stress, which disrupts protein folding and triggers apoptosis in malignant cells. DIM Powder also plays a role in modifying estrogen metabolism and functions as an antagonist to both estrogen and androgen receptors, including a strong inhibitory effect on the pure androgen receptor (AR). This makes it of particular interest in hormone-dependent cancers such as prostate and breast cancer.

 

In Vivo Studies

In animal studies, DIM Powder continues to demonstrate significant biological activity, particularly in its role as a radioprotective compound. When administered repeatedly, DIM has been shown to protect against the damaging effects of systemic radiation. This protective effect is attributed to its ability to activate the ATM pathway in normal tissues, reinforcing the body's natural defense mechanisms at the cellular level.

Oral administration of DIM in mice, typically at a dose of 250 mg/kg, has shown high bioavailability without signs of acute toxicity, supporting its potential for therapeutic use. These findings highlight DIM's safety profile and its effectiveness in reaching target tissues through convenient routes of administration.
