Professor David Sinclair is a well-known NMN scientist in the field at Harvard Medical School. He said in an interview that he takes NMN bulk powder and resveratrol powder every morning. Why does NMN bulk powder need resveratrol powder as a "gasoline"?
We know that all the functions of NMN are realized by converting to NAD+ in vivo. And NMN is a direct precursor of NAD+, which means that only one step is needed to convert NMN to NAD+ in vivo. This step requires the participation of a conversion enzyme nicotinamide mononucleotide adenyltransferase.
After the conversion of NMN to NAD+, NAD+ will participate in a variety of biochemical reactions in the body, helping to repair DNA damage and activate the Sirtuins family of longevity proteins to achieve a range of functions that delay aging.
Moreover, NAD+ is not excreted from the organs like vitamin C, but has its own circulatory system, which recycles the products of NAD+ use (mostly nicotinamide NAM) into NAD+ again. It is also the main pathway for NAD+ synthesis in vivo.
During the process of NAD+ recycling, it is regulated by the activity of another conversion enzyme, NAMPT (nicotinamide phosphoribosyltransferase). In animals, NAMPT is divided into two types according to its location: extracellular (e.g. in the blood) NAMPT called eNAMPT and intracellular NAMPT called iNAMPT.

It is easy to see that NMNAT is an important "activator" for the conversion of NMN into NAD+, while NAMPT determines whether NAM bulk powder can be reused and regulates the homeostatic level of NAD+, so to speak. This is why a single formulation of NMN is only effective for a short period of time, but without the regulation of these two conversion enzymes, this recycling "production line" will soon be in a negative state.
How does NMN bulk powder and resveratrol powder to activate this recycling channel?

A research team from Macau University of Science and Technology recently published their findings in Pharmacology Research & Perspectives, in which they investigated the effects of NMN and selected dietary supplements on NAD+ levels, including resveratrol, and discovered that the combination of NMN increased NAD+ levels in specific tissues and organs.
The researchers used male mice aged 6-8 weeks and divided them into four groups: (1) control, (2) NMN, (3) NMN and resveratrol combination, with the control group receiving PBS (phosphate buffer) orally twice and the NMN group receiving PBS (phosphate buffer) orally once followed by NMN (500 mg/kg). In the combined group, resveratrol (50 mg/kg) was given orally first, followed by NMN (500 mg/kg) orally.
Combination improves the distribution of NMN bulk powder in tissues
Although NMN is rapidly metabolized in plasma (within 30 minutes), transformation in tissues usually takes longer. Researchers measured NMN concentrations in different tissues 1, 2, 4 and 6 hours after oral administration, as shown in Figure 1.
After NMN alone, NMN was mainly enriched in the liver. In contrast, after co-administration with resveratrol, the concentration of NMN in the liver decreased and the level in the heart increased by 1.39-fold. It indicates that NMN co-administration can improve the distribution of NMN among tissues.
Combination improves the distribution of NR in tissues
Why should NR be measured?
In fact, NMN and NR are closely related. Because NMN is too large, it cannot enter the cell directly and must be transported. First, NMN is dephosphorylated by CD73 to generate NR, which then enters the cell through the equilibrium nucleoside transport protein. Subsequently, NR is reconverted to NMN via the cytoplasmic NRK enzyme.
As we can see in Figure 2, the concentration of NR is generally low in brain, lung and muscle tissues in either group, with higher concentrations present only in the kidney, which may be related to the high expression of NRK and CD73 in kidney tissues. Compared with the NMN group, the resveratrol coadministration group showed significantly higher NR concentrations in cardiac tissues, which were 1.93 times higher than before. In contrast, NR levels in brain, kidney and liver were significantly higher in the ginsenoside Rg3 and Rh2 co-administration group. This result was very close to the previous distribution of NMN, indicating that the level of NMN is indeed inseparable from NR.
Combination can increase the NAD+ level in some tissues
It is well known that NMN acts as a direct precursor of NAD+. Oral administration of NMN alone significantly increased NAD+ levels in liver and kidney, indicating that NMN supplementation improved the energy metabolism levels in liver and kidney tissues. Surprisingly, the combination of NMN and resveratrol increased cardiac and muscle NAD+ levels by 1.59-fold and 1.72-fold, respectively, compared to NMN bulk powder alone. The elevated NAD+ levels in the heart reduced cardiac hypertrophy and protected the heart, and the elevated NAD+ levels in the muscle reduced muscle damage, inflammation and fibrosis.
