Our testing center test by UV detector. Its wavelength 206nm, Hypersil silica liquid chromatography column, column length 250mm. internal diameter 4.6mm, particle size 5um. column temperature 35 degrees C, air flow rate 2.5L/min, flow rate 1ml/min, injection 20μl. mobile phase: acetonitrile-methanol-phosphoric acid mixed solution (95:5:1)

From the graph, we can see that the peak height response value is low. And the baseline separation is not reached. The baseline is uneven and uneven. And 206nm is the terminal absorption. The detection result is unstable. It directly leads to 40% of the PS detection result of 70%.

Under the above chromatographic conditions, the instrument was stabilized and the baseline was smooth, the control solution was injected into 10ul and 20ul respectively. And the relative standard deviation (RSD) of peak area was ≤3.0%. The sample solution was injected into 10ul.

What Are The Pure Phosphatidylserine PS Testing Of Problems Encountered?

In the process of daily testing, it was found that the sunflower phosphatidylserine powder in the common solvent (referring to the conventional PS solvent chloroform: methanol = 2:1) is not completely dissolved, the solution is turbid and opaque state. The effect on the detection content results is large, and the separation of impurity peaks is low.

What Are The Phosphatidylserine PS Testing Of The Solution?

Under the same chromatographic conditions, we adjusted the solvent for the problem of incomplete dissolution of sunflower PS, from the original chloroform:methanol (2:1) to the original chloroform:methanol (2:1).

The original chloroform: methanol (2:1) was replaced by hexane: isopropanol: water = 3:2:1, mixed and left to stratify, and the upper layer was taken as the sunflower PS solvent.

The new solvent test experiment, the adjusted solvent, the sunflower PS effect is remarkable, the solution is clear and translucent, high separation of impurity peaks, more accurate content detection.

Therefore, in the sunflower PS test to the new solvent test results prevail.

 

We compare our results in Method 2 with the methods of foreign third-party testing organizations:

20% phosphatidylserine PS and 50% phosphatidylserine PS

In the detection of 20% PS and 50% PS, the error of method 2 and foreign third-party institutions 31P-NMR method is within 1%. The assay docking was good.
