Testing Method:

Here is a general method for analyzing ginseng sample extract powder using HPLC and UV spectroscopy:

Materials:

- HPLC system with UV detection

- Column: C18, 150 mm x 4.6 mm, 5 μm

- Mobile phase: acetonitrile and water (50:50)

- Ginseng extract powder

- Methanol

- Water

- UV spectrophotometer

- Quartz cuvette

Instructions:

1. Prepare a standard stock solution of ginsenosides (or other active components) in methanol.

2. Dilute the stock solution with methanol to prepare working standard solutions of different concentrations.

3. Prepare ginseng samples extract powder by dissolving in methanol and filter through a 0.45 μm membrane filter.

4. Inject the standard solutions and sample solutions onto the HPLC column and run the gradient elution program for about 30 minutes.

5. Record the UV spectra of the eluted peaks and compare with the standard reference spectra to identify the components.

6. Quantify the components by measuring the peak area at the wavelength with the maximum absorbance.

7. Calculate the content of each component in the ginseng extract sample powder based on the calibration curve of the corresponding standard solutions.

8. Verify the results by repeating the analysis and calculating the relative standard deviation (RSD).
