The Test Method 

Method of determination

First, instruments and reagents

1. Instrument: UV-2101 PC UV-Vis Spectrophotometer

2. Reagents:

2% NH4Fe(SO4)2•12H2O 2N hydrochloric acid aqueous solution: Measure 17 mL of concentrated hydrochloric acid, add water to 100 mL, add 2 g of NH4Fe(SO4)2•12H2O, shake and dissolve, and obtain.

Acid n-butanol solution: Add 5 mL of concentrated hydrochloric acid to 95 mL of n-butanol and mix.

 

Second, solution preparation

1. Preparation of organic cranberry extract powder sample solution Accurately weigh about 100mg of pine bark extract sample, place it in a 50mL volumetric flask, add methanol to dissolve and make up. Accurately draw 2mL into a 50mL volumetric flask and dilute to the mark with methanol, which is the test solution. Accurately draw 1 mL of the test solution into a 10 mL stoppered test tube, add 0.2 mL of 2N NH4Fe(SO4)2•12H2O in 2N hydrochloric acid aqueous solution, and 6 mL of acidic n-butanol solution, tighten the sealing cap, shake well, and loosen the sealing cap. The reaction was heated in a 95 ° C water bath for 60 min, taken out, rapidly cooled in an ice water bath, and placed at room temperature, and diluted to 10 mL with acidic n-butanol to obtain a sample solution to be tested.

3. Sample blank solution preparation Replace the sample with methanol and treat it as sample solution, ie

Third, sample determination

Turn on the organic cranberry extract powder power of the instrument and preheat it for 20 minutes. After the instrument passes the self-test, the absorbance of the standard solution and the sample solution is determined by using a solution of n-butanol hydrochloride as a blank at 546 nm.
