Assay of Sesamin in Black Sesame Seed Extract

1. Reagents

Water: HPLC grade

Methanol: HPLC grade

Standard: ChromaDex, purity of 98%

 

2. Equipment

High performance liquid chromatography with UV detector.

 

3. Chromatographic system

Column: 4.6 mm × 150 mm, 5 μm, C-18

Mobile phase: Methanol: Water (80: 20, V/V)

Column temperature: Room temperature

Flow rate: 0.5 mL/min

Injection size: 20 μL

Wavelength: 287 nm

 

4. Procedure

4.1 Preparation of Standard Solution

Dissolve 10 mg of Black Sesame Seed Extract Sesamin Standard in 40 mL of 95% ethanol solution. Sonicate for 30 min to dissolve. Cool to room temperature, dilute with 95% ethanol to 50 mL and mix well. Pass through a filter of 0.45 μm or finer pore size.

4.2 Preparation of Sample Solution

Dissolve 10 mg of Sesame Extract powder in 40 mL of 95% ethanol solution. Sonicate for 30 min to dissolve. Cool to room temperature, dilute with 95% ethanol to 50 mL and mix well. Pass through a filter of 0.45 μm or finer pore size.

4.3 Test

Separately inject the test solution and standard solution into the chromatograph, and record the chromatograms. Measure the peak areas of the Sesamin peak.

 

5. Calculation

Calculate the percentage of Sesamin in the sample by the formulas as following:
