● Solubility
Marigold flower extract lutein is insoluble in water and propylene glycol. It is slightly soluble in oil, hexane, acetone, dichloromethane, and ethanol. And lutein powder bulk is soluble in ethyl acetate, tetrahydrofuran, chloroform, etc. Its stability in the solvent is anhydrous ethanol & GT, Ethyl acetate & GT, Tetrahydrofuran & GT; Toluene.

 

● Stability
The stability of bulk lutein is poor, and it is mainly affected by oxygen, light, heat, metal ions, pH, and other factors. For example, the isomerization reaction of lutein during heat treatment can produce 9-cis and 13-cis lutein. Therefore, in preservation, lutein crystal pure products or materials containing lutein should be sealed in a vacuum or filled with inert gas packaging to avoid light and low-temperature preservation.

3.1. Weighing 40mg, place the bulk Lutein sample in 50 ml volumetric flask, add solvent extract, dissolve,constant volume by Solvent Extract, shake,stand; take 1ml solution in 50 ml volumetric flask, constant volume by ethanol, shake,stand, test.

3.2. Measure the absorbance in 446±1 nm on the UV Spectrophotometer (ethanol as a blank), record the max absorbance A;

4.5.1. Preparation of standard solution:

Weighing 10mg of standards bulk lutein, place the sample in 50 ml volumetric flask, add mobile phase, dissolve,constant volume by mobile phase, shake.

4.5.2. Preparation of sample solution

Weighing 25mg sample wholesale lutein powder bulk, place the sample in 100 ml volumetric flask, add mobile phase, dissolve,constant volume by mobile phase, shake.

 

4.6. Sample testing

Accurately inject the standard solution when the chromatographic system is stabilized under the condition. Separately inject equal volumes of standard solution and sample solution to the chromatograph, record the chromatograms. repeat twice,obtain the area percentage of Lutein (P1)and the area percentage of zeaxanthin(P2).

 

4.7. Calculating
