Luteolin is a naturally occurring flavonoid. Its chemical name is 3', 4', 5, 7-tetrahydroxyflavone. It its widely distributed in the plant kingdom. For example, it is present in acacia, peanut shells, honeysuckle, wild chrysanthemum and broccoli. Luteolin powder can be extracted from honeysuckle. The chemical structure formula is shown in Figure 1.
Luteolin has a high penetration capacity in the skin. It can reach deep into the skin layer. The effects are as follows: antioxidant, delayed aging, anti-inflammatory, whitening, antibacterial, inhibition and elimination of skin discoloration. However, it suffers from the important problem of poor solubility, which leads to the limitation of Luteolin powder application in cosmetics. Its solubility and utilization can be improved by solid dispersions, cyclodextrin inclusion complexes, phospholipid complexes, nanoemulsions, polymer micelles and other formulation techniques. Advances in Formulation Research
01 Solid dispersion
Solid dispersion (SD) is a solid dispersion obtained by combining a refractory ingredient with a highly soluble carrier by fusion (fusion) method, solvent method or molten solvent method. This results in drug release and greatly increases the dissolution of insoluble drugs. The solubility of luteolin powder in luteolin solid dispersion (Lut-SD) and luteolin phospholipid complex solid dispersion (Lut-PC-SD) prepared by Xiangtao Deng et al. was increased from (61.09±0.09) µg/mL to 6-7 times of the original. The solubility comparison is shown in Figure 3.

02 Cyclodextrin inclusion complexes
Cyclodextrin is a cyclic oligosaccharide. It has a lipophilic inner cavity and a hydrophilic outer surface, and is able to interact with a variety of guest molecules to form non-covalent inclusion complexes. It is mainly used to enhance the water solubility, physicochemical stability and bioavailability of drugs. Xudong Wang et al. prepared sustained-release antimicrobial nanofibers (PVA-Lut-IC-NF) by incorporating luteolin powder inclusion complex (Lut-IC) into poly(vinyl alcohol) (PVA) by electrostatic spinning.PVA-Lut-IC-NF has good water solubility and thermal stability. And it has significant inhibitory effect on Staphylococcus aureus, Candida albicans and Escherichia coli.
03 Phospholipid complexes
Phospholipid complex is a self-assembly of liposome-like vesicles formed in water by interactions between phospholipids and active ingredients through non-covalent bonding (charge transfer, hydrogen bonding, van der Waals forces, etc.). It increases the stability of the active and prolongs the duration of action. Related studies have shown that phospholipid complexes containing lignans. The solubility in water is about 2.5 times that of pure luteolin powder.
04 Nanoemulsion
Nanoemulsion is made of two immiscible liquids. With the help of appropriate surfactant and co-surfactant composition of the particle size between 1 ~ 100 nm homogeneous dispersion system. It has the advantages of increasing the solubility of the active substance and improving the bioavailability, etc. Shin et al. prepared luteolin nanoemulsions consisting of poly(ethylene oxide)-block poly(ε-caprolactone) and lecithin polymerized at the O/W interface. It showed excellent stability during repeated freeze-thaw cycles and long-term storage. And minoxidil was used as a positive control to apply it to the depilated skin of mice, and the results showed similar hair growth after 17 days. However, the hair on the surface of the skin treated with luteolin powder nanoemulsion was more uniform and soft.
05 Polymeric micelles
Polymer micelles are amphiphilic block copolymers that self-assemble in water into ordered aggregates with a core/shell structure. The hydrophobic chains of the block copolymer form the core of the micelle. They are used to encapsulate hydrophobic substances. The hydrophilic chains form its outer shell and maintain the spatial stability of the micelle. It was shown that luteolin powder was assembled into methoxy polyethylene glycol polycaprolactone (mPEG5K-PCL10K) micelles. It not only has a high stability and encapsulation rate, but the water solubility of luteolin is also greatly improved.
